A Rapid Genetic Assay for the Identification of the Most Common Pocillopora damicornis Genetic Lineages on the Great Barrier Reef

چکیده
Pocillopora damicornis (Linnaeus, 1758; Scleractinia, Pocilloporidae) has recently been found to comprise at least five distinct genetic lineages in Eastern Australia, some of which likely represent cryptic species. Due to similar and plastic gross morphology of these lineages, field identification is often difficult. Here we present a quick, cost effective genetic assay as well as three novel microsatellite markers that distinguish the two most common lineages found on the Great Barrier Reef. The assay is based on PCR amplification of two regions within the mitochondrial putative control region, which show consistent and easily identifiable fragment size differences for the two genetic lineages after Alu1 restriction enzyme digestion of the amplicons.

Broadcast Spawning by Pocillopora Species on the Great Barrier Reef
The coral genus Pocillopora is one of the few to include some species that broadcast spawn gametes and some species that brood larvae, although reports of reproductive mode and timing vary within and among species across their range. Notably, the ubiquitous Pocillopora damicornis has been described as both a brooder and spawner, although evidence of broadcast spawning is rare. Here, we report o...
متن کاملPopulation Genetics of an Ecosystem-Defining Reef Coral Pocillopora damicornis in the Tropical Eastern Pacific
BACKGROUND Coral reefs in the Tropical Eastern Pacific (TEP) are amongst the most peripheral and geographically isolated in the world. This isolation has shaped the biology of TEP organisms and lead to the formation of numerous endemic species. For example, the coral Pocillopora damicornis is a minor reef-builder elsewhere in the Indo-West Pacific, but is the dominant reef-building coral in the...
متن کاملMixed asexual and sexual reproduction in the Indo-Pacific reef coral Pocillopora damicornis
Pocillopora damicornis is one of the best studied reef-building corals, yet it's somewhat unique reproductive strategy remains poorly understood. Genetic studies indicate that P. damicornis larvae are produced almost exclusively parthenogenetically, and yet population genetic surveys suggest frequent sexual reproduction. Using microsatellite data from over 580 larvae from 13 colonies, we demons...
متن کاملWhere do clonal coral larvae go? Adult genotypic diversity conflicts with reproductive effort in the brooding coral Pocillopora damicornis
Earlier studies of the coral Pocillopora damicornis provide a conflicting picture of its use of sexual and asexual reproduction for population maintenance. In Western Australia, colonies are asexually viviparous, and populations appear to be maintained by localised asexual recruitment but founded by genotypically diverse colonists. However, on Australia’s Great Barrier Reef (GBR), as in many ot...

Background & Objectives: Zagros Mountains, which are between the flatlands of Mesopotamia and the Iranian plateau, surround the western border of the country. This region was the cradle of great civilizations like Mesopotamia and Elam. This study aimed to test the boundary role of the Zagros Mountains in the genetic structure of the region through evaluating the paternal lineages inheritance ...
